Identification and functional analysis of endogenous nitric oxide in a filamentous fungus

چکیده
In spite of its prevalence in animals and plants, endogenous nitric oxide (NO) has been rarely reported in fungi. We present here our observations on production of intracellular NO and its possible roles during development of Neurospora crassa, a model filamentous fungus. Intracellular NO was detected in hypha 8-16 hours after incubation in Vogel's minimal liquid media and conidiophores during conidiation using a fluorescent indicator (DAF-FM diacetate). Treatment with cPTIO, an NO scavenger, significantly reduced fluorescence levels and hindered hyphal growth in liquid media and conidiation, whereas exogenous NO enhanced hyphal extension on VM agar media and conidia formation. NO scavenging also dramatically diminished transcription of con-10 and con-13, genes preferentially expressed during conidiation. Our results suggest that intracellular NO is generated in young hypha growing in submerged culture and during conidia development and regulate mycelial development and conidia formation.

Response to hypoxia, reduction of electron acceptors, and subsequent survival by filamentous fungi.
Filamentous fungi usually inhabit normoxic environments by utilizing oxygen as a substrate for respiration and for the biosynthesis of some essential cellular components. This review examines the metabolic mechanisms used by filamentous fungi under oxygen-limited (hypoxic) conditions. Denitrification is one mechanism through which Fusarium oxysporum and other fungi reduce nitrate or nitrite to ...
متن کاملEffect of endogenous nitric oxide on cardiac ischemic preconditioning in rat
Introduction: Ischemic Preconditioning (IPC) is the phenomen that happens on the heart by one or several short periods of ischemia followed by reperfusion that improve the postischemic recovery of mechanical function. Ischemic preconditioning (IPC) may protect the heart from ischemia reperfusion injury by nitric oxide formation. This study investigated the effect of ischemic preconditioni...
متن کاملNitric oxide ameliorates salinity tolerance in Pyrodwarf pear (Pyrus communis) rootstocks by regulating polyamine content
Nitric oxide (NO), an endogenous signaling molecule, is involved in various physiological processes and stress responses in plants. In the present research, Pyrodwarf pear (Pyrus communis) rootstocks were grown by nutrient solution to investigate the effects of sodium nitroprusside (SNP) application as an NO donor at 0, 0.1, 0.5, and 1 mM levels on plant stress tolerance, content of ma...
متن کاملFIVE ALPHA DIHYDROTESTOSTERONE (5α-DHT) MAY MODULATE NITRIC OXIDE RELEASE VIA ENDOGENOU S CYTOKINES IN PERITONEAL MA CROPHA GES OF NZB/BALBc MICE
Recent studies have established that sex hormones directly or indirectly affect T and B cells and macrophages by manipulating the production of cytokines. In this study the possibility of the effect of 5a-DHT on macrophage (MΦ) nitric oxide (NO) release via interleukin-l, 6 (lL-1β, IL-6) or tumor necrosis factor-a (TNFα) was investigated. The endogenous cytokines IL-1β, IL-6 and TNF-α were ...
متن کاملNephrotoxicity of Isosorbide Dinitrate and Cholestasis in Rat: The Possible Role of Nitric Oxide
Background: Nitric oxide (NO), a major chemical form of endothelium-derived relaxing factor and an important regulator of vascular tone, is released by endothelial cells. The role of NO is not restricted to the vascular system, and it participates in the regulation of renal hemodynamics and renal excretory function. There are increasing evidences indicating that the elevated levels of NO play a...
